Quantification

For accurate quantitative analysis of TLC / HPTLC separations, a scanner is used to perform densitometric evaluation, i.e. produce chromatograms from bands measured at the appropriate wavelengths.
Different modes may be required to obtain accurate and consistent results, such as absorption or fluorescence.
Calibration can be performed on pre-defined standard samples, making it possible to qualify and quantify unknown samples.
These instruments offer a wide range of possibilities:
-Recording of adsorption spectra at desired points to facilitate substance identification
Measurements using reference wavelengths (two-wavelength measurements) to reduce background noise.
-Multi-wavelength analysis, to detect the most suitable measurement wavelengths for chromatograms of unknown samples.
